چکیده

Microspheres made of high refractive index melamine resin are shown to enhance the fluorescence from single molecules in solution by seven-fold, and simultaneously reduce the observation volume by thirteen-fold, as compared to state-of-the-art confocal microscopy. This fluorescence enhancement is demonstrated to dramatically increase the signal-to-noise ratio in fluorescence correlation spectroscopy and reduce the experiment integration time by fifty-fold. We also provide the first description of dual-color fluorescence cross-correlation spectroscopy (FCCS) enhanced by a dielectric microsphere, and report comparable enhancement factors as for the single color case.

Two-photon fluorescence correlation spectroscopy with high count rates and low background using dielectric microspheres

Two-photon excitation fluorescence is a powerful technique commonly used for biological imaging. However, the low absorption cross section of this non-linear process is a critical issue for performing biomolecular spectroscopy at the single molecule level. Enhancing the two-photon fluorescence signal would greatly improve the effectiveness of this technique, yet current methods struggle with me...

Optical-fiber-microsphere for remote fluorescence correlation spectroscopy.

Fluorescence correlation spectroscopy (FCS) is a versatile method that would greatly benefit to remote optical-fiber fluorescence sensors. However, the current state-of-the-art struggles with high background and low detection sensitivities that prevent the extension of fiber-based FCS down to the single-molecule level. Here we report the use of an optical fiber combined with a latex microsphere...

Disposable microscope objective lenses for fluorescence correlation spectroscopy using latex microspheres.

We explore the combination of a latex microsphere with a low NA lens to form a high performance optical system, and enable the detection of single molecules by fluorescence correlation spectroscopy (FCS). Viable FCS experiments at concentrations 1-1000 nM with different objectives costing less than $40 are demonstrated. This offers a simple and low-cost alternative to the conventional complex m...

Single molecule detection of DNA looping by NgoMIV restriction endonuclease.

Single molecule fluorescence resonance energy transfer (FRET) and fluorescence correlation spectroscopy were used to investigate DNA looping by NgoMIV restriction endonuclease. Using a linear double-stranded DNA (dsDNA) molecule labeled with a fluorescence donor molecule, Cy3, and fluorescence acceptor molecule, Cy5, and by varying the concentration of NgoMIV endonuclease from 0 to 3 x 10(-6) M...

Tracking-FCS: Fluorescence correlation spectroscopy of individual particles.

We exploit recent advances in single-particle tracking to perform fluorescence correlation spectroscopy on individual fluorescent particles, in contrast to traditional methods that build up statistics over a sequence of many measurements. By rapidly scanning the focus of an excitation laser in a circular pattern, demodulating the measured fluorescence, and feeding these results back to a piezoe...
